EP02.01-002. Development of Circulating and Tissue Biomarkers Predicting Immune Phenotype and Response to Immunotherapy in NSCLC.

Immunotherapy, specifically immune checkpoint inhibitors, has significantly improved the treatment of non-small cell lung cancer (NSCLC). However, there is a need for biomarkers that can predict patient response to immunotherapy. Recent research has shown that certain circular RNAs (circRNAs) associated with checkpoint receptors could serve as potential biomarkers of cancer and immune phenotype. This study aimed to detect these circRNAs in the blood of NSCLC patients using a simple RT-PCR method.<br /><br />Plasma samples were collected from ten NSCLC patients and purified for circulating free circRNA. The circRNA expression was then analyzed using custom-designed primers and agarose gel electrophoresis. The study found that specific circRNAs associated with checkpoint receptors were detected in the plasma of some patients. For example, one patient showed the presence of hsa_circ_0128427 associated with TIM-3 expression, along with other circRNAs. Another patient showed the presence of B7-H3, a marker associated with tumor invasion and poor prognosis.<br /><br />While this study is not conclusive, it suggests that circRNAs associated with checkpoint receptors can be detected in the plasma of NSCLC patients using simple RT-PCR. This opens the possibility of using more sensitive and specific methods to detect these biomarkers. Furthermore, these circRNAs could serve as minimally invasive markers to guide and monitor immune checkpoint therapy.<br /><br />Overall, this study highlights the potential of circRNAs as biomarkers for predicting immune phenotype and response to immunotherapy in NSCLC. Further research using more sophisticated detection methods is needed to validate these findings and potentially improve patient stratification and treatment outcomes.
