The researchers also demonstrated that a STAT3 inhibitor upregulated the mRNA and protein expressions of ERRFI1, while a STAT3 activator downregulated its expression. LC-C enhanced the transcriptional activity of the ERRFI1 promoter, which was further enhanced by the STAT3 inhibitor and reduced by the STAT3 activator. IL-6, a cytokine that activates STAT3, attenuated the effects of LC-C on ERRFI1 expression and promoter transcription activity.<br /><br />Furthermore, LC-C effectively inhibited the repair of DNA damage caused by ionizing radiation and increased the radiosensitivity of the radiation-resistant lung cancer cells. On the other hand, IL-6 attenuated these effects of LC-C on DNA damage repair and radiosensitivity.<br /><br />In conclusion, the phosphorylation of STAT3 plays a crucial role in the regulation of ERRFI1 expression and radiosensitivity in lung cancer treated with LC-C. Phosphorylated STAT3 affects ERRFI1 expression by regulating the transcriptional activity of its promoter. These findings provide insights into the mechanism of action of LC-C in enhancing the radiosensitivity of lung cancer cells and suggest its potential use as a therapeutic agent for lung cancer treatment.
