EP07.02-002. Investigating the Role of MicroRNAs in Cell Growth and Cisplatin-Sensitivity of Malignant Pleural Mesothelioma

A study conducted by researchers at the University Hospital Zurich in Switzerland explored the role of microRNAs in the growth of malignant pleural mesothelioma (PM) cells and their sensitivity to the standard chemotherapy regimen of platinum/pemetrexed. The researchers identified several microRNAs that not only had prognostic and predictive biomarker potential, but also showed therapeutic potential. Overexpression of miRs-380-5p, -221-3p, -30a, -210, -625-3p, miR-222, and miR-19b led to reduced PM cell growth but had no effect on non-malignant cells. Additionally, overexpression of miR-210, -625-3p, -19b, and -222 also significantly reduced colony forming ability in the PM cell lines. Furthermore, overexpression of miR-380-5p, miR-221-5p, miR-625-3p, and miR-19b sensitized some cell lines to cisplatin, but not to pemetrexed. The researchers observed that the response to the cisplatin-pemetrexed doublet was in line with the sensitization towards cisplatin. The study also assessed the effect of microRNA overexpression on the response to the cisplatin-pemetrexed doublet and found that while overexpression of the tested microRNAs did not sensitize cells to pemetrexed, it did sensitize cells to cisplatin alone and the doublet treatment to varying degrees. Overall, the data suggest that several microRNAs play a role in regulating PM cell growth and response to chemotherapy, with microRNA overexpression having a sensitizing effect on cisplatin. This study provides valuable insights into the potential therapeutic use of microRNAs for treating PM.
