EP08.01-009. TRBV Haplotype Profile Was Related to Immune-Induced Adverse Events in Chinese Patients With Advanced NSCLC

Researchers from Shanghai conducted a study to investigate the relationship between T-cell receptor beta variable (TRBV) haplotypes and immune-induced adverse events (irAEs) in Chinese patients with advanced non-small cell lung cancer (NSCLC) who received immunotherapy. Previous studies have shown that TRBV gene variants are associated with irAEs in Caucasian NSCLC patients, but this relationship had not been explored in Chinese patients before.<br /><br />The study analyzed TRBV allele profiles in 143 Chinese NSCLC patients who received either first- or second-line immunotherapy. The patients were divided into six TRBV haplotype groups based on the results of TCR sequencing. The study found that TRBV haplotype group 1 was associated with a higher risk of developing irAEs. The proportion of patients with grade II irAEs was significantly higher in group 1 compared to other groups.<br /><br />Additionally, the study found that the number of TRBV genes and the presence of uncommon V alleles were higher in group 1, suggesting a potential mechanism for the increased occurrence of irAEs in this group. Furthermore, in a subgroup of 47 patients, there was a higher mutation frequency in the Wnt signaling pathway in patients from TRBV haplotype group 1 compared to other groups.<br /><br />These findings suggest that TRBV haplotype profile may be related to the development of irAEs in Chinese NSCLC patients receiving immunotherapy. Understanding this relationship could help identify patients who would be at a higher risk of experiencing irAEs and may require closer monitoring or tailored treatment approaches to minimize adverse events.<br /><br />Further research is warranted to validate these findings and explore the underlying mechanisms of the association between TRBV haplotypes and irAEs. Nonetheless, this study provides valuable insights into personalized immunotherapy approaches in the Chinese NSCLC population.
