Pdf Summary
In this study, the authors aimed to explore the landscape and dynamic changes of peripheral immune cells in non-small cell lung cancer (NSCLC) patients receiving immunotherapy. They also aimed to identify potential biomarkers that could reflect response to immunotherapy.

They collected peripheral blood mononuclear cells (PBMCs) from 33 NSCLC patients before treatment and after 2 cycles of immunotherapy. They analyzed the PBMCs using full spectrum flow cytometry to assess the composition and frequencies of various immune cell populations, including CD4 T cells, CD8 T cells, regulatory T cells, T helper cells, B cells, dendritic cells, natural killer cells, monocytes, and myeloid-derived suppressor cells.

The results showed that NSCLC patients who experienced less than nine months of progression-free survival had a significant reduction in the frequency of CD4 effector memory T cells after treatment. On the other hand, NSCLC patients who achieved at least nine months of progression-free survival had a decreased frequency of CD8 effector memory T cells after treatment. Reduction in the frequency of PD-1 memory T cells was also associated with a higher likelihood of achieving longer progression-free survival.

Furthermore, there was a significant increase in the proportion of myeloid dendritic cells in patients with shorter progression-free survival, but not in those with longer progression-free survival.

Overall, these findings suggest that the landscape and dynamic changes of peripheral immune cells can potentially serve as biomarkers to predict response to immunotherapy in NSCLC patients. Further investigation is needed to validate these findings and explore their clinical implications.
