EP16.01-022. A Signature of B Cells-Related Gene Pairs Predicts Oncologic Outcomes and Response to Immunotherapy in NSCLC Patients

Researchers from the Shandong Cancer Hospital and Institute in China have developed a prognostic model that can predict oncologic outcomes and response to immunotherapy in non-small-cell lung cancer (NSCLC) patients. The model is based on a novel signature of 23 B cell-related gene pairs (BRGPs) comprising 28 unique B cell-related genes (BRGs), identified through the analysis of public single-cell RNA sequencing data. The researchers utilized the Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) datasets to construct the BRGPs model, which showed significant power in distinguishing good or poor prognosis in NSCLC patients and was verified using the GSE31210 dataset.<br /><br />The prognostic accuracy of the model was further confirmed through univariate and multivariate cox regression analysis in the GEO cohorts. The risk model was found to be associated with sex, TNM stage, immune score, tumor purity, and various tumor-infiltrating immune cells. Additionally, the model was found to be positively correlated with tumor mutational burden (TMB) score, indicating a potential role in guiding immune checkpoint inhibitor treatment.<br /><br />Further analysis revealed that the low-risk group enriched several immune-related pathways, while the high-risk group enriched proliferation-related pathways. The study also demonstrated a positive correlation between the risk score and TMB score, suggesting that NSCLC patients with a high-risk score may have higher TMB and could potentially benefit from immune checkpoint blockade therapy.<br /><br />Overall, this novel BRGPs model could help assess the prognosis of NSCLC patients and guide immunotherapy treatment decisions in clinical practice. The study highlights the importance of considering the complex interactions between B cell-related genes for a more accurate prognostic model.
