EP16.01-028. Immunomodulatory Effects of Cryoablation Combined With Immune Checkpoint Inhibitors in a Murine Lung Cancer Model

This study investigated the immunomodulatory effects of cryoablation combined with immune checkpoint inhibitors (ICIs) in a murine lung cancer model. The researchers focused on myeloid-derived suppressor cells (MDSCs) and T-cell immunity, which play a role in the tumor microenvironment. <br /><br />Results showed that the combination of cryoablation and ICI had synergistic anti-tumor effects. Tumor growth was suppressed in the ICI groups compared to the control group, but there was no significant difference between anti-PD-1 and anti-PD-L1 groups. However, cryoablation combined with ICIs further suppressed tumor growth, with significant differences observed in the Cryoablationanti-PD-1 and Cryoablationanti-PD-L1 groups compared to the Cryoablation group.<br /><br />Flow cytometry analysis showed that PMN-MDSCs in the bone marrow were significantly decreased in the Cryoablationanti-PD-1 and Cryoablationanti-PD-L1 groups compared to the Cryoablation group. Additionally, CD8-positive effector memory T cells in the tumor were increased in the Cryoablationanti-PD-1 and Cryoablationanti-PD-L1 groups compared to the Cryoablation group.<br /><br />The researchers concluded that the combination of cryoablation and ICI had synergistic anti-tumor effects, which could be evaluated by monitoring MDSCs and T-cell immunity. These findings suggest that cryoablation combined with ICIs may disrupt the immunosuppressive tumor microenvironment and enhance immune response.<br /><br />Overall, this study provides evidence for the potential of combining cryoablation with immune checkpoint inhibitors as a therapeutic approach for lung cancer, with implications for improving immune response and suppressing tumor growth.
