EP16.03-017. Spatial Heterogeneity of Tumor Microenvironment of Lung Adenocarcinoma Associated With Genomic Alterations

Researchers conducted a study to analyze the spatial heterogeneity of tumor microenvironment (TME) in lung adenocarcinoma and its association with gene alterations. They used spatial transcriptomics, a recently developed method that allows for the analysis of gene expression levels with spatial information. The goal was to uncover the complex spatial network within the TME, which could provide insights into cancer evolution and potential therapeutic implications.<br /><br />The study included six samples from two patients with advanced lung adenocarcinoma. Spatial transcriptomic data was produced using Visium, a technology from 10X Genomics. Unsupervised clustering analysis revealed 13 distinct clusters, with significant variation in the proportion of clusters between patients and samples. <br /><br />The researchers also conducted RNA-seq analysis and network analysis to explore the transcriptomic-genomic alterations in multiple regions within the tumor. They found that the spatial distribution of cell types within the tumor showed mixed patterns in some regions and distinct immune cell infiltration in others. They also performed laser capture microdissection and whole-genome sequencing in six regions of one patient to assess spatial genomic alterations. Interestingly, they found no significant differences in copy number alterations among the regions, suggesting a lack of branched evolution.<br /><br />Overall, this study provides valuable insights into the spatial heterogeneity of the TME in lung adenocarcinoma. By employing spatial transcriptomics and genomic analysis, the researchers were able to uncover the complex network between immune-stromal cells within the tumor. These findings have potential implications for understanding cancer progression and developing targeted therapies.
