EP16.03-039. Characterizing SHPRH as a Novel Tumor Suppressor Gene in Lung Adenocarcinoma

Researchers from the Department of Integrative Oncology at the BC Cancer Research Institute in Canada have identified a novel tumor suppressor gene, SHPRH, in lung adenocarcinoma. The team performed whole exome sequencing on never smoker lung adenocarcinoma tumors and identified SHPRH as having frequent mutation, copy number deletion, and biallelic inactivation.<br /><br />Further analysis revealed that SHPRH is frequently lost in lung adenocarcinoma and is associated with poorer survival outcomes. The researchers found that low SHPRH expression is associated with poor overall survival and progression-free survival in all lung adenocarcinoma patients.<br /><br />To evaluate the effect of altered SHPRH expression on the development and progression of lung adenocarcinoma, the team conducted in vitro and in vivo tumorigenicity assays. They found that re-expression of SHPRH in SHPRH-inactivated lung adenocarcinoma cell lines reduced their colony formation and tumor burden.<br /><br />Based on these findings, the researchers concluded that SHPRH may function as a tumor suppressor in lung adenocarcinoma. They also suggested that further research should be conducted to explore the mechanism by which SHPRH operates as a tumor suppressor and evaluate its cooperative effect with DNA damaging agents/carcinogens on genomic instability and lung tumorigenesis.<br /><br />The discovery of SHPRH as a novel tumor suppressor gene in lung adenocarcinoma has significant implications for early screening and treatment of the disease. SHPRH may become an important clinical biomarker for identifying individuals with an increased risk for lung cancer and predicting their therapeutic response.
