EP16.03-046. Tumor Grade Associated Genomic Mutations in Chinese Patients with Lung Cancer

This study aimed to identify tumor grade-associated genomic mutations in Chinese patients with lung cancer. Tumor samples were collected from patients and analyzed using next-generation sequencing targeting a panel of pan-cancer genes. The sequencing depth was over 1000 to improve mutation detection sensitivity. The study compared gene mutations in low-grade, middle-grade, and high-grade tumors using Fisher's exact test and inferred enriched pathways in each tumor grade.<br /><br />Lung cancer is a significant cause of cancer-related deaths in China, and high-grade lung cancer is particularly dangerous. Understanding the mechanisms of high-grade lung cancer could help develop new treatments for these patients. The study found that high-grade tumors in lung cancer patients were more affected by bacteria, such as the Epstein-Barr virus, compared to low-grade tumors. The tumors from these patients specifically showed mutations in TP53 and PIK3CA genes, which could serve as biomarkers for these patients. These findings underscore the importance of developing tailored treatments for patients with these mutations.<br /><br />The study included 173 Chinese patients with lung cancer, with a higher proportion of female patients. Most patients were at the early stage of the disease, and the median tumor mutation burden was 2.5. When comparing high-grade tumors to low-grade tumors, a higher percentage of mutations in TP53 and PIK3CA genes were found in high-grade tumors. Pathway analysis revealed that high-grade tumors were enriched with mutations in bacterial invasion of epithelial cells, Epstein-Barr virus infection, and the Wnt signaling pathway.<br /><br />Overall, this study provides insights into tumor grade-associated genomic mutations in Chinese lung cancer patients. The findings suggest that high-grade tumors may be influenced by bacteria and highlight the potential use of TP53 and PIK3CA mutations as biomarkers for these patients. These findings support the need for personalized treatment approaches for patients with these specific mutations.
