EP16.04-007. ALDH2 Downregulation, Aberrant DNA Methylation, and Loss of Stemness Pathways as an Enhanced Biomarker in Lung Adenocarcinoma

The study investigates the role of Aldehyde Dehydrogenase 2 (ALDH2) in lung adenocarcinoma (LUAD), the most common type of lung cancer. The researchers conducted a meta-analysis of ALDH2 gene expression and examined its association with overall survival (OS), protein expression, and DNA methylation in LUAD patients. They also analyzed the signaling pathways and gene-gene interactions involving ALDH2 in LUAD.<br /><br />The results showed that ALDH2 was downregulated in LUAD, both at the gene and protein expression levels. The downregulation of ALDH2 was strongly associated with poorer overall survival in LUAD patients. The study also found significant differences in DNA methylation levels of ALDH2 between LUAD cancer and normal tissues, with specific methylation patterns correlating with the survival time of LUAD patients.<br /><br />Furthermore, the downregulation of ALDH2 was observed in the proteomic expression profile, particularly in stemness pathways, which play a crucial role in regulating the balance between stemness and differentiation in adult stem cells. The study also identified correlations between ALDH2 and the activities of immunological B cells and CD4 T cells.<br /><br />Overall, the findings suggest that the downregulation of ALDH2, aberrant DNA methylation, and deficiency of stemness signaling pathways could serve as potential prognosis factors and therapeutic targets in LUAD.<br /><br />This research was conducted by an international Ph.D. program for Cell Therapy and Regeneration Medicine, as well as master programs in Clinical Genomics and Proteomics, and Artificial Intelligence in Medicine, all affiliated with Taipei Medical University in Taiwan. Funding for the study was provided by the Ministry of Science and Technology of Taiwan.
