EP16.04-009. The Proliferative Effect of 27-Hydroxycholesterol as a Selective Estrogen Receptor Modulator on Pathology of NSCLC

Researchers from Tokyo Medical University in Japan conducted a study to investigate the role of 27-hydroxycholesterol (27HC) in promoting the proliferation of non-small cell lung cancer (NSCLC) cells. 27HC is an oxysterol that has been shown to promote the growth of breast cancer cells as a selective estrogen receptor modulator (SERM). The study aimed to determine if 27HC acts similarly in lung cancer cells, which also express estrogen receptors.<br /><br />In the study, tumor and nontumor regions were collected from NSCLC patients who underwent surgical lung resection. The researchers found that the level of 27HC was significantly higher in the tumor region compared to the nontumor region, particularly in Grade III tumors.<br /><br />In vitro experiments using an ERβ-positive lung cancer cell line (H23) were also conducted. The results showed that exposure to 27HC promoted the proliferation of lung cancer cells, similar to estradiol (E2), a natural estrogen hormone. This effect was blocked when an ER antagonist was used, suggesting that 27HC exerts its proliferative effect through estrogen receptors.<br /><br />Immunohistochemical staining of lung tissue samples further supported these findings. The expression of ERβ and the mRNA levels of its target gene cMYC, as well as tumor-promoting factors HIF1a and VEGF, were significantly higher in the tumor region compared to the nontumor region.<br /><br />Based on these results, the researchers concluded that 27HC, produced by alveolar macrophages in the lung, promotes the proliferation of NSCLC cells expressing estrogen receptors and worsens the pathological conditions of the tumors. They suggest that targeting 27HC could be a potential strategy for cancer therapy in NSCLC.
