EP16.04-028. PHLPP2 Regulates Ferroptosis Through Nrf2 Pathway to Affected Cell Cycle and Apoptosisin Lung Squamous Cell Carcinoma

This study aimed to investigate the role of the PH domain and leucine-rich repeat protein phosphatase 2 (PHLPP2) in lung squamous cell carcinoma, particularly in relation to the nuclear factor erythrocyte-derived 2-like 2 (Nrf2) pathway. Lung cancer is a leading cause of mortality worldwide, and PHLPP2 has been identified as a tumor suppressor in various cancers. Previous research has shown that PHLPP2 inhibits the Nrf2 response, which regulates the expression of antioxidant proteins.<br /><br />The study analyzed data from the GEO and TCGA databases and found that PHLPP2 expression was lower in tumors compared to normal tissues, while Nrf2 expression was higher. Additionally, a negative correlation was observed between PHLPP2 and Nrf2 in lung squamous cell carcinoma. Flow cytometry analysis revealed that inhibiting PHLPP2 expression led to a decrease in the G1/S ratio and a decrease in apoptotic cells. Western blotting showed that knocking down PHLPP2 increased the expression of p-Nrf2 and HO-1, while PTGS2 expression decreased.<br /><br />Based on these findings, the study concludes that after PHLPP2 knockdown in lung cancer cells, the cell cycle was activated, apoptosis was inhibited, and ferroptosis (a type of regulated cell death) was also inhibited. These results suggest that PHLPP2 may regulate ferroptosis and apoptosis through the Nrf2 pathway in lung squamous cell carcinoma.<br /><br />In summary, PHLPP2 appears to play a role in the regulation of cell cycle, apoptosis, and ferroptosis in lung squamous cell carcinoma. Understanding the mechanisms involved in these processes could potentially lead to the development of new therapeutic strategies for the treatment of this type of lung cancer.
