PP01.57 (Poster) Understanding the molecular mechanism of Supinoxin on Small cell Lung Cancer Therapy

Researchers at Purdue University have been studying the molecular mechanism of Supinoxin, a potential treatment for small cell lung cancer (SCLC). They focused on the role of the RNA helicase DDX5 in SCLC, as it is overexpressed in this type of cancer. The stability of DDX5 in SCLC lines was found to be attributed to post-translational modification (phosphorylation). The researchers are investigating the repurposing of a small molecule believed to target DDX5 for SCLC treatment. This molecule has shown growth inhibition in multiple cancer cell types and has been successful in inhibiting tumor growth and improving survival in mouse models of triple-negative breast cancer. Treatment with Supinoxin demonstrated inhibition of proliferation and colony formation in SCLC cell lines, as well as inhibition of tumor growth and improved survival in mice with SCLC tumors. However, the precise mechanism of action of Supinoxin remains unknown, as it did not lead to significant changes in the levels or localization of c-Myc, DDX5, and β-Catenin proteins. Additionally, Supinoxin was found to induce downregulation of mitochondrial functions. The researchers have conducted Western blot analysis and phosphoproteome analysis to further understand the effects of Supinoxin treatment. Small cell lung cancer is a highly aggressive cancer type with fast growth, rapid metastasis, and chemoresistance, and effective treatment options are currently lacking. The researchers hope that their findings will contribute to the development of new approaches to SCLC therapy.
