P1.20. Prognostic and Predictive Relevance of 3q Amplification in Squamous Cell Carcinoma of the Lung

Researchers conducted an analysis of 476 cases of squamous cell carcinoma of the lung (LUSC) to evaluate the prognostic and predictive relevance of 3q amplification (AMP) in this type of cancer. They found that 3q AMP is the most common genetic alteration in LUSC, with the most frequent amplified region being 3q26-3q28. However, 3q AMP is not commonly identified in another type of lung cancer called LUAD. The study identified a 1.7MB minimal common region (MCR) of AMP within a 17.5MB amplified region on 3q. All 25 genes within the MCR were frequently amplified, with mRNA overexpression observed in some of the genes. The researchers also analyzed the correlation of MCR status with the tumor immunophenotype using multiple RNA-based algorithmic scores. <br /><br />The analysis showed that MCR AMP was detected in 46.4% of the LUSC cases. It was strongly associated with PIK3CA AMP and CDKN2A homozygous deletion. Furthermore, MCR AMP was found to have prognostic value in LUSC, with better disease-specific survival (DSS) and progression-free interval (PFI) observed in cases with MCR AMP. MCR AMP was also associated with higher levels of PD-L1, suggesting that it could predict for a higher response to PD-L1 inhibitors.<br /><br />The study also investigated the presence of tumor-infiltrating cells and immune checkpoints in relation to MCR AMP. There was a trend towards a higher prevalence of immuno-inhibitory cells in MCR non-amplified cases, while PD-L1 expression was higher in MCR-amplified cases.<br /><br />The researchers plan to validate these results in a retrospective racially diverse cohort with advanced LUSC using a fluorescent in situ hybridization (FISH) probe to detect MCR AMP. Overall, the study provides insights into the prognostic and predictive relevance of 3q AMP in squamous cell carcinoma of the lung, specifically the minimal common region of AMP within 3q.
