EP01.01. Association between ACEI-Induced Cough and the Risk of Lung Cancer: A Mendelian Randomization Study - PDF(Slides)

The authors of this study aimed to examine the association between the use of angiotensin-converting enzyme inhibitors (ACEIs) and the risk of lung cancer using a Mendelian randomization (MR) approach. Observational studies and meta-analyses have shown conflicting results on this association, and it is unknown whether ACEI-induced cough is related to lung cancer development. The authors collected single nucleotide polymorphisms (SNPs) that mimic the effect of ACEIs and ACEI-induced cough and conducted a meta-analysis of existing genome-wide association studies for various lung cancers. The relationship was quantified using inverse variance weighting, weighted median, and MR-Egger methods.<br /><br />The results of the analysis showed a significant association between ACE inhibition and lung cancer development, specifically small cell lung cancer in Europeans. Additionally, ACEI-induced cough was associated with a higher risk of lung cancer in Europeans, but not in Asians. It was also found that ACEI-induced cough was associated with lung adenocarcinoma. The authors suggest that patients with hypertension who experience dry cough as a side effect of ACEI use should consider switching to an alternative antihypertensive treatment.<br /><br />The findings of this study contribute to the understanding of the relationship between ACEI use, ACEI-induced cough, and the risk of lung cancer. However, it is important to note that the study has limitations, and further research is needed to confirm these associations. Overall, this study provides evidence supporting the need for careful consideration of the potential risks and benefits of ACEI use, particularly in individuals at risk for lung cancer.
