EP01.01. Casual Effects of Some Chronic Respiratory Diseases on Squamous Cell Lung Cancer: A Mendelian Randomization Study - PDF(Abstract)

This study aimed to investigate the causal relationship between chronic respiratory diseases (such as asthma, chronic obstructive pulmonary disease, idiopathic pulmonary fibrosis, obstructive sleep apnea, and bronchiectasis) and squamous cell lung cancer. The researchers used a method called mendelian randomization (MR), which utilizes genetic data to analyze causal effects.<br /><br />The study selected instrumental variables for the respiratory diseases from large genome-wide association studies. These variables were genetically associated with the diseases and were statistically significant. Data on squamous cell lung cancer were collected from the International Lung Cancer Consortium.<br /><br />The primary analysis, using the inverse variance weighting (IVW) method for two-sample MR, showed a positive association between genetically predicted chronic obstructive pulmonary disease (COPD) and idiopathic pulmonary fibrosis (IPF) with squamous cell lung cancer. However, no significant association was found between asthma, obstructive sleep apnea, and bronchiectasis with squamous cell lung cancer.<br /><br />Additional analyses adjusted for smoking and exposure to tobacco were also performed. The results showed that COPD remained a significant risk factor for squamous cell lung cancer, while the effects of IPF became non-significant. The association between IPF and smoking and exposure to tobacco was also not statistically significant.<br /><br />Based on their findings, the researchers concluded that COPD and IPF are independent risk factors for the development of squamous cell lung cancer. They suggested that further research, both epidemiological and mechanistic, should be conducted to explore the interactions between these respiratory diseases and lung cancer.
