In this scientific inquiry, the authors sought to understand the role of DEPDC1 in lung adenocarcinoma (LUAD). They analyzed the expression patterns of DEPDC1 in LUAD and normal tissues and found that DEPDC1 expression was significantly higher in LUAD tissues. High DEPDC1 expression was also associated with poor prognosis in LUAD patients.<br /><br />The authors conducted in vitro experiments to investigate the effects of DEPDC1 on the proliferation of LUAD cells. They found that down-regulation of DEPDC1 inhibited the proliferation, migration, and invasion ability of LUAD cells. Furthermore, they observed a modulation of the mTOR pathway upon down-regulation of DEPDC1, specifically affecting the expression of mTOR pathway markers and their phosphorylation levels.<br /><br />Moreover, the authors identified a novel regulatory axis where DEPDC1 regulates the growth of LUAD cells via KIF2C. They found that KIF2C expression was significantly higher in LUAD tissues and was also associated with poor prognosis in LUAD patients. The inhibitory effect of down-regulated DEPDC1 on LUAD cell proliferation, invasion, and migration was alleviated by overexpression of KIF2C.<br /><br />In conclusion, DEPDC1 overexpression is associated with poor prognosis in LUAD patients. DEPDC1 may affect the mTOR pathway through its regulation of KIF2C, thereby influencing the proliferation, invasion, and migration of LUAD cells. These findings highlight the significant role of DEPDC1 in LUAD tumorigenesis and provide potential targets for future therapeutic interventions.
