EP02.01. Suppression of Telomerase RNA Component Induces Autophagy and Ferroptosis in Lung Cancer via Regulation of AMP-actived Protein Kinase - PDF(Abstract)

This document presents the findings of a study conducted to investigate the role of the telomerase RNA component (TERC) in lung adenocarcinoma (LUAD) progression. Long non-coding RNAs (lncRNAs) are known to play a role in tumorigenesis, and TERC is a lncRNA that is involved in the addition of telomere repeats. However, the specific effects of TERC dysregulation in LUAD are not well understood.<br /><br />The researchers compared the expression levels of TERC in cancerous and adjacent normal lung tissues using RNA-seq analysis. They found that TERC expression was upregulated in lung cancer tissues. To further understand the role of TERC, the researchers performed functional assays in LUAD cell lines by using siRNA-mediated knockdown of TERC. They observed that the knockdown of TERC inhibited the migration and invasion of LUAD cells.<br /><br />The researchers also investigated the underlying mechanisms of TERC in LUAD progression. They found that silencing TERC increased the expression of autophagy-related proteins LC3B, Beclin-1, and AMP-activated protein kinase (AMPK). Additionally, the expression of p62 protein and ferroptosis-regulated proteins GPX4 and SLC7A11 were diminished. Importantly, they found that the inhibition of AMPK counterbalanced the effects of TERC knockdown on autophagy and ferroptosis in LUAD cells.<br /><br />Overall, the findings of this study suggest that the suppression of TERC in lung cancer promotes autophagy and ferroptosis by regulating AMPK. These findings provide insights into the role of TERC in tumorigenesis and may have clinical significance in understanding the dysregulation of TERC in lung cancer. The study highlights potential molecular therapeutic targets for lung cancer treatment.
