This study aimed to investigate the expression profile of circular RNAs (circRNAs) in lung adenocarcinoma (LUAD). The researchers used second-generation sequencing to analyze 14 pairs of postoperative LUAD specimens, including cancer tissues and adjacent tissues. They detected 5,242 distinct circRNAs and found aberrantly expressed circRNAs in the cancerous tissues compared to the normal tissues. <br /><br />The study identified a circular RNA-mediated competing endogenous RNA (ceRNA) network in human LUAD. The ceRNA network refers to the regulatory interactions between circRNAs, microRNAs (miRNAs), and their target mRNAs. The researchers constructed a circRNA and miRNA binding network to understand these interactions.<br /><br />The researchers also found that certain circRNAs, such as hsa_circ_0120106, has_circ_0007342, has_circ_0005937, and circRNA_0001640, were significantly upregulated in LUAD cases compared to healthy controls. They performed ROC curve analysis, which showed the potential of these circRNAs as diagnostic markers for LUAD.<br /><br />Additionally, the study conducted a KEGG analysis to explore the signaling pathways associated with the target genes of the dysregulated circRNAs. The results revealed several significantly enriched signaling pathways.<br /><br />Moreover, the researchers identified a ceRNA network that regulates LUAD. This network consists of interactions between circRNAs, miRNAs, and mRNAs. They also developed a lasso logistic model using the coefficients of circRNAs to predict the diagnosis of non-small cell lung cancer (NSCLC).<br /><br />In summary, this study provided insights into the expression profile and potential diagnostic significance of circRNAs in LUAD. The findings contribute to the understanding of circRNAs' role in cancer and suggest their potential as biomarkers for LUAD diagnosis.
