EP03.03. Single-nucleus RNA Sequencing Dissecting the Tumor Microenvironment of Liver Metastasis of Non-small Cell Lung Cancer - PDF(Abstract)

This study aimed to investigate the tumor microenvironment of liver metastasis in non-small cell lung cancer (NSCLC) using single-nucleus RNA sequencing. Liver metastasis is a critical factor in the survival of NSCLC patients and affects the efficacy of various treatments, including immunotherapy. However, the underlying mechanisms and complexity of the liver tumor microenvironment remain unclear. <br /><br />The researchers performed single-nucleus RNA sequencing on needle biopsy specimens of liver metastasis from three NSCLC patients. A total of 47,339 cells were sequenced and analyzed. Additionally, single-nucleus RNA sequencing data from three healthy liver tissues were obtained from a database for comparison. The analysis revealed the variation in gene expression profiles of stromal and immune cells in the microenvironment.<br /><br />The results identified 17 clusters, with clusters 16 and 17 exclusively found in liver metastasis. Compared to healthy liver tissues, liver metastasis samples had more epithelial cells (including tumor cells) but fewer hepatocytes. Surprisingly, more Kupffer cells, endothelial cells, T cells, and B cells were found in liver metastasis samples. Several genes, including PRRC2B, HEPACAM2, KIT, and MMP9, were significantly enriched in liver metastasis samples. Notably, the study also found differences in gene expression between liver metastasis and healthy liver tissues within specific cell types.<br /><br />Further analysis revealed enriched pathways in different cell types, such as ribosome, complement, and coagulation cascades in Kupffer cells, endocytosis in T cells, and adherens junctions in B cells in the liver metastasis microenvironment.<br /><br />In conclusion, this study provides insights into the transcriptome of the metastatic liver tumor microenvironment in NSCLC at a single-cell resolution. The findings shed light on potential mechanisms of liver metastasis in NSCLC, offering new perspectives for understanding and targeting this aspect of the disease.
