EP03.05. Aberrant Methylation of ZNF577 As a Potential Prognostic Biomarker for Lung Adenocarcinoma - PDF(Abstract)

The study focused on investigating the methylation pattern of ZNF577 in lung adenocarcinoma, a type of lung cancer. The researchers examined DNA methylation using bisulfite pyrosequencing and analyzed mRNA expression levels of ZNF577 using RT-PCR. They also evaluated ZNF577 tissue expression using immunohistochemistry. <br /><br />The results showed that the mRNA expression levels of ZNF577 were significantly higher in normal tissues compared to lung adenocarcinoma tissues. Additionally, ZNF577 mRNA expression was higher in females, non-smokers, and stage I samples. The overall survival and disease-free survival rates were significantly higher in the group with high ZNF577 mRNA expression compared to the low expression group. The DNA methylation levels of ZNF577 were significantly higher in lung adenocarcinoma tissues compared to normal tissues, and it was inversely related to mRNA expression. The methylation levels were lower in lepidic samples compared to papillary samples, and higher in lung adenocarcinoma with vascular invasion. <br /><br />Out of the examined lung adenocarcinoma samples, 44 were positive for ZNF577 tissue expression and had better survival rates compared to the negative group. However, the difference in overall survival and disease-free survival between the negative and positive samples was not statistically significant. Cox regression analysis showed better survival rates in younger patients without pleural invasion. <br /><br />The study suggests that ZNF577 could be a potential prognostic biomarker for lung adenocarcinoma. The findings highlight the importance of DNA methylation patterns in cancer and provide insights into the functional role of ZNF577 in lung adenocarcinoma.
