A study was conducted to characterize the genomic landscape of non-small cell lung cancer (NSCLC) in the Republic of Ireland. The researchers obtained NSCLC patient samples referred to the Cancer Molecular Diagnostics Lab at St James’s Hospital and used the Oncomine Focus Assay to analyze molecular alterations in 35 hotspot genes, 23 gene fusions, and 19 copy number variants. They collected basic clinical and demographic data and performed a retrospective search of Ion Reporter software for genomic data analysis.<br /><br />The study included 2,052 NSCLC patients with a median age of 69 years. Fifty-three percent of the patients were male, and 47% were female. The patients were referred from 23 different hospital sites across all four provinces of Ireland. The researchers found that females were more likely to have clinically significant alterations, including KRAS and EGFR mutations. Actionable alterations were more frequently detected in younger patients aged 50 years or younger. They also observed that fusions involving ALK, ROS1, and RET were present in 14.7% of cases in the younger age group, compared to only 3% in older patients.<br /><br />The study identified several common driver oncogenes in NSCLC patients. The most prevalent genomic aberration was wildtype, followed by KRAS mutations, EGFR mutations, fusions (ALK/ROS1/RET/FGFR3), BRAF mutations, MET exon 14 skip, HER2/ERBB2 mutations, PIK3CA mutations, and others. Co-alterations, such as EGFR/PIK3CA, KRAS/PIK3CA, and KRAS/IDH1, were also observed.<br /><br />This study provides insight into the genomic landscape of NSCLC in the Irish population. The findings suggest that gender and age may influence the presence of specific genomic alterations in NSCLC patients. These results contribute to a better understanding of the molecular characteristics of NSCLC in Ireland and may have implications for personalized treatment approaches for NSCLC patients in the future.
