EP06.03. HER3 Expression in Archived Tissue Samples from Patients with NSCLC Across Various Genomic Subtypes and Characteristics - PDF(Abstract)

This study aimed to characterize HER3 expression and its association with other tumor biomarkers in patients with non-small cell lung cancer (NSCLC) across different genomic subtypes. The study used archival tissue samples from patients with advanced or metastatic NSCLC, including those with specific genetic mutations. HER3 expression was evaluated through immunohistochemistry using an assay developed by Ventana Medical Systems. The study included samples from 199 patients, with 98% showing HER3 membrane expression. The mean membrane H-score was 131. Among the evaluated samples, 10% had EGFR-activating mutations, all of which exhibited HER3 membrane expression. Similar observations were made for samples with EGFR exon 20 insertions, ALK rearrangements, and KRAS G12C mutations. In samples with undetermined driver oncogene status, HER3 membrane expression was widely observed. In patient samples with PD-L1 protein expression, detectable membrane HER3 expression was found in 98-100% of samples. The study concluded that HER3 membrane expression was observed across all included genomic subtypes of NSCLC, both in patients who had received systemic anticancer therapy and those who had not. These findings support previous studies suggesting that targeting HER3 could be beneficial in patients with diverse subtypes of NSCLC. Overall, the study highlights the potential of HER3 as a promising biomarker and therapeutic target in NSCLC.
