EP06.03. HER3 Expression in Archived Tissue Samples from Patients with NSCLC Across Various Genomic Subtypes and Characteristics - PDF(Slides)

A study was conducted to evaluate the expression of the membrane HER3 protein in non-small cell lung cancer (NSCLC) tumors. The study found that membrane HER3 was expressed in 98.5% of the NSCLC tumor samples, regardless of the genomic subtype or prior treatment with systemic anticancer therapy. This suggests that targeting HER3 could be effective for a broad range of NSCLC patients.<br /><br />Previous studies have shown that HER3 plays a role in activating signaling pathways associated with oncogenesis, proliferation, migration, and metastasis in NSCLC. It has also been linked to poor prognosis in malignant solid tumors. The objective of this study was to further characterize HER3 expression and its associations with clinical and tumor characteristics.<br /><br />The study evaluated HER3 expression in archived tissue samples from 203 patients with advanced or metastatic NSCLC. The expression was assessed using immunohistochemistry. It was found that 98.5% of the samples showed membrane HER3 expression, and the mean membrane H-score was 129.55.<br /><br />The study also assessed the presence of genomic alterations and PD-L1 expression in the tumor samples. It was observed that all samples with EGFR-activating mutations exhibited membrane HER3 expression. Other mutations reported included EGFR exon 20 insertions, ALK rearrangements, and KRAS G12C mutations.<br /><br />In conclusion, this study confirms previous findings that HER3 is highly expressed in NSCLC tumors and may be a promising target for new therapies. The results suggest that targeting HER3 could be effective across different subtypes of NSCLC and regardless of prior treatment with systemic anticancer therapy. Further research is needed to investigate the potential therapeutic implications of targeting HER3 in NSCLC.
