EP13.01. Single-Cell RNA Sequencing Reveals Immune Microenvironment of Small Cell Lung Cancer-associated Malignant Pleural Effusion - PDF(Abstract)

This research examined the immune microenvironment of small cell lung cancer (SCLC) in patients with malignant pleural effusion (MPE) using single-cell RNA sequencing. The study aimed to explore the features of disseminated tumor cells in liquid biopsy for SCLC, an area with limited data available. The researchers obtained pleural effusion samples from a patient with SCLC-MPE and performed single-cell transcriptome sequencing to build a gene library. They then used bioinformatics analysis to reveal the characteristics of the tumor immune microenvironment.<br /><br />The results showed that a total of 8,008 high-quality cells were obtained and divided into 10 cell clusters. These clusters were identified as B cells, T cells, myeloid cells, NK cells, and cancer cells. Further subgroup analysis revealed different subtypes within B and T cells, including translational B cells, memory B cells, naive B cells, plasma B cells, as well as naive CD8 T cells, CD8 effector_GZMK T cells, CD8 effector_GNLY T cells, naive CD4 T cells, CD4 effector T cells, CD4 memory T cells, and Treg cells. The researchers also found the expression of SCLC-related genes during the differentiation of T and B cells.<br /><br />Cell communication analysis showed a complex interaction network between T cells, B cells, and tumor cells. The interaction between these immune cells and tumor cells primarily occurred through MHC II, IL-16, GALECTIN, and APP signaling pathways. The study concluded by highlighting the need for further research to understand the relationship between these pathways and the efficacy of immunotherapy for SCLC.<br /><br />Overall, this research provides valuable insights into the immune microenvironment of SCLC-associated MPE, shedding light on potential targets for immunotherapy in these patients.
