Researchers at Guangdong Provincial People’s Hospital in China have developed a nanosystem for the delivery of two drugs, all-trans retinoic acid (ATRA) and gefitinib, to overcome resistance to Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitors (EGFR-TKIs) in lung adenocarcinoma. EGFR-TKIs are commonly used to treat lung cancer, but some patients develop resistance, leading to tumor progression and metastasis. The researchers utilized a polymer called polyphenylalanine (Polymer 8P4) to encapsulate ATRA and gefitinib, creating Gef/ATRA@8P4-NPs.<br /><br />In vitro and in vivo experiments were conducted to investigate the efficacy and safety of the nanosystem. The Gef/ATRA@8P4-NPs exhibited uniform and spherical nanoparticles, and they demonstrated sustained-release behavior of the drugs in a simulated tumor acidic microenvironment. When tested on EGFR-TKI resistant lung cancer cells, the nanosystem showed potent cytotoxicity and induced apoptosis.<br /><br />In animal models, the Gef/ATRA@8P4-NPs demonstrated superior therapeutic efficacy and better biosafety compared to control groups. Tumor volume and weight were significantly reduced, and no hepatic metastasis foci were observed. The researchers believe that this nanosystem could be a promising strategy for the treatment of advanced metastatic lung adenocarcinoma.<br /><br />The study was supported by grants from the Natural Science Foundation of Guangdong and the International Science and Technology Cooperation Program of Guangdong. The research findings provide a new approach to addressing EGFR-TKI resistance and improving outcomes for lung cancer patients. Further studies and clinical trials are needed to validate the effectiveness and safety of the polyphenylalanine nanosystem in human patients.
