Pdf Summary
A study presented at the WCLC 2023 conference titled "APC-1, A Novel Aptamer-Cisplatin Conjugate, Exhibits Promising Anti-tumor Effect in Lung Cancer" discusses a new treatment approach for lung cancer using an aptamer-cisplatin conjugate called APC-1. Cisplatin, a commonly used chemotherapy drug, has limited effectiveness against leptomeningeal metastasis (LM) and is associated with neurotoxicity.

The researchers developed APC-1 by conjugating an aptamer with cisplatin. Aptamers are synthetic DNA or RNA oligonucleotides that can target specific molecules. In this case, the aptamer was designed to target lung cancer cells.

The study used mouse models to evaluate the efficacy of APC-1. Results showed that APC-1 had a superior tumor suppressive effect compared to cisplatin when administered at an equivalent dose in the lung cancer LM models. In the subcutaneous xenograft models, APC-1 had an identical tumor suppressive effect to cisplatin. Importantly, mice treated with APC-1 did not experience significant weight loss, unlike those treated with cisplatin.

The aptamer sequence of APC-1 was detectable in both the plasma and cerebrospinal fluid, suggesting that APC-1 can cross the blood-brain barrier. This finding indicates the potential of APC-1 in treating lung cancer LM.

In conclusion, the study demonstrates that APC-1 has a superior tumor suppressive effect at lower systemic cisplatin concentrations compared to cisplatin alone. The ability of APC-1 to penetrate the blood-brain barrier suggests a potential role in treating lung cancer with leptomeningeal metastasis. This research highlights the development of a novel treatment approach for lung cancer using an aptamer-cisplatin conjugate.
