P1.12. CD66c Activates EGFR Signalling Pathway and Promotes EMT in Lung Adenocarcinoma - PDF(Abstract)

The study used transcriptome sequencing on patient-derived LUAD organoids and found that the carcinoembryonic antigen-related cell adhesion molecule (CEACAM) family, particularly CD66c, was commonly upregulated in LUAD. <br /><br />The researchers performed quantitative real-time PCR (QPCR) and Western blotting to confirm the overexpression of CD66c in primary LUAD tumors. They also found that CD66c expression was significantly correlated with lymph node positive status. Further experiments showed that knockdown of CD66c suppressed the migration and metastasis ability of LUAD cells. The knockdown of CD66c also led to a decrease in the expression of epithelial-mesenchymal transition (EMT) markers, indicating its role in EMT modulation. <br /><br />Additionally, the researchers observed that stable overexpression of CD66c enhanced the migratory capability of LUAD cells and increased the expression of EMT markers. Mechanistically, they found that CD66c activated the EGFR signaling pathway by promoting the phosphorylation of EGFR and binding to the extracellular domain of EGFR and its binding partner LGALS3. <br /><br />Overall, these findings suggest that CD66c plays a critical role in promoting EMT and migration in LUAD cells through its activation of the EGFR signaling pathway. The researchers concluded that CD66c could be a promising therapeutic target for intervention in LUAD.
