Researchers from Nanjing Medical University and Shanghai Chest Hospital in China have developed a blood-based DNA methylation panel for the early detection of lung cancer (LC). LC is the leading cause of cancer-related mortality worldwide, and the prognosis of LC is strongly influenced by the stage at initial diagnosis. The 5-year overall survival rate drops from 85% for stage IA to 6% for stage IV. Therefore, it is important to develop a tool for early detection of LC patients.<br /><br />The study used mass spectrometry to process blood samples from a training-validation set of LC cases and controls. The researchers established a five-gene panel for the early detection of LC using this approach. The blood samples from LC cases were collected before surgery and any tumor-related treatment.<br /><br />The results of the study showed that the blood-based DNA methylation panel has the potential to detect LC even at Stage I. The MALDI-TOF mass spectrometry platform was found to be efficient and stable for the semi-quantitative determination of DNA methylation signatures.<br /><br />In terms of performance, the blood-based DNA methylation panel had an AUC (area under the curve) of 0.87 for distinguishing LC cases from controls in the training set, and an AUC of 0.84 in the validation set. The panel also showed good sensitivity and specificity in differentiating LC cases from controls at different stages, including stage IA and stage I.<br /><br />In conclusion, the blood-based DNA methylation panel developed in this study shows promise for the early detection of LC, even at an early stage. The panel could potentially improve the prognosis and survival rate of LC patients by enabling early detection and intervention. Further studies and validation are needed to confirm the findings.
