P1.22. Comparison of NGS and FISH Platforms to Test MET Amplification in Chinese Lung Cancer Patients: A Prospective Study - PDF(Slides)

The study aimed to compare the effectiveness of Next-Generation Sequencing (NGS) and Fluorescence in Situ Hybridization (FISH) in detecting MET amplification in Chinese lung cancer patients. MET amplification is important to understand resistance mechanisms to EGFR-Tyrosine kinase inhibitors. The sensitivity of MET amplification detection in circulating tumor DNA (ctDNA) by NGS was also evaluated.<br /><br />Tissue biopsies were performed on patients with progressive disease resistant to EGFR-TKIs. MET amplification was tested using both NGS and FISH on the tissue samples. Blood samples were also taken and ctDNA was analyzed for MET amplification using NGS. The concordance of MET amplification detection between tissue NGS and FISH, as well as ctDNA NGS and FISH, was calculated.<br /><br />The study enrolled 67 patients resistant to EGFR-TKI therapies, with 24 resistant to 1st- or 2nd-generation TKIs and 43 resistant to 3rd-generation TKIs. The detection rate of MET amplification in patients resistant to 3rd-generation TKIs was significantly higher (41.9%) compared to those resistant to 1st- and 2nd-generation TKIs (16.7%).<br /><br />The results showed that tissue NGS had good concordance with FISH in detecting MET amplification in post EGFR-TKI resistance NSCLC. However, the sensitivity of ctDNA NGS in detecting MET amplification needs further optimization and improvement.<br /><br />The study was supported by AstraZeneca Investment (China) Co., Ltd.<br /><br />In conclusion, NGS showed good concordance with FISH in detecting MET amplification in tissue samples from lung cancer patients. However, the sensitivity of ctDNA NGS for MET amplification detection needs to be improved. Further research is required to optimize ctDNA NGS as a diagnostic tool.
