P1.22. Identification of MIF as a Multiple Primary Lung Cancer Related Gene Based on RNA Sequencing Technology and Study of Its Function - PDF(Abstract)

This study aimed to explore the transcriptome characteristics and related genes of multiple primary lung cancer (MPLC) through bulk and single-cell RNA sequencing analysis. Tumor tissues and paired paracancerous tissues from 18 MPLC patients were collected and compared with data from lung adenocarcinoma patients in the Cancer Genome Atlas (TCGA) database. The cell composition of MPLC tissues was described using multicenter source single-cell sequencing data. Differentially expressed genes (DEGs) and marker genes of various subtypes of epithelial cells were used to identify MPLC-related genes. The expression and impact of these genes on the proliferation, migration, and invasion abilities in lung-derived cell lines were further explored. <br /><br />The study identified 1553 DEGs between MPLC and single lung adenocarcinoma and found that MPLC may have more inhibitory immune cell infiltration. Single-cell sequencing analysis revealed significant heterogeneity among different foci in MPLC, with epithelial cells being subdivided into 18 subtypes. The study identified macrophage migration inhibitory factor (MIF) as an MPLC-related gene, and the MIF-CD74 pathway was found to be widely involved in the communication between different cell types of MPLC. Overexpression of MIF was associated with poor prognosis in lung adenocarcinoma, and cell experiments showed that MIF may induce carcinogenesis in some AT2 cells. <br /><br />In conclusion, this study identified MIF as a gene associated with multiple primary lung cancer and found that it may have a promoting effect on lung cancer. The findings provide a new approach for the diagnosis and treatment of multiple primary lung cancer.
