They identified seven major cell types, with myeloid cells and T cells being the most abundant. The expression of FOLR2 was significantly higher in IA compared to earlier stages. The infiltration of NR4A3 and FOXP3 positive cells also increased during LUAD progression. The researchers hypothesized that FOLR2TAM elevation in IA might be partially due to the recruitment of DCs and other TAMs through chemokine recruitment.<br /><br />They further suggested that FOLR2TAM might collaborate with DCs to recruit NR4A3CD4T cells, which could potentially convert to regulatory CD4 T lymphocytes (Tregs) in the progression of LUAD. Pseudotime analysis supported the idea that NR4A3CD4T cells may differentiate into Tregs, as they expressed Treg differentiation-related genes. The study provided a single-cell transcriptome atlas of LUAD, highlighting the high cellular heterogeneity and transcriptomic features at different pathological stages.<br /><br />In conclusion, the study revealed a potential trajectory involving FOLR2TAM, DCs, NR4A3CD4T cells, and Tregs in shaping the immunosuppressive microenvironment in invasive LUAD. This understanding may have implications for developing targeted therapies to disrupt this trajectory and enhance immune responses against LUAD.
