Pdf Summary
The presentation titled "Restoring Lymphocytes Using NKTR-255 after Chemoradiotherapy in Solid Tumors (RESCUE)" at the WCLC 2023 conference discusses the use of NKTR-255, a novel clinical stage polymer-conjugated interleukin-15 (IL-15) agonist, to restore lymphocyte counts after chemoradiation (CRT) in patients with locally advanced non-small cell lung cancer (NSCLC). Radiation induced lymphopenia (RIL) is a common side effect of radiation therapy that can lead to worse clinical outcomes and reduce the effectiveness of immunotherapy. NKTR-255 is designed to improve the proliferation and survival of T cells by engaging the IL-15 receptor complex.

The study aims to recruit 30 patients with locally advanced NSCLC who have received standard CRT followed by one year of durvalumab (standard of care). The primary objectives of the study are to monitor the safety of NKTR-255 after CRT and estimate the restoration of lymphocyte counts after its administration. Exploratory objectives include characterizing the pharmacokinetics, immunogenicity, and immune profile changes associated with NKTR-255.

The patients will receive NKTR-255 intravenously in conjunction with durvalumab. Blood samples will be collected to assess lymphocyte counts before each dose and at specific intervals. PK samples will also be collected before and after NKTR-255 infusion. Safety assessments will be conducted after each infusion, and dose de-escalation is permitted if necessary.

The study's early stopping rules include safety measures to control the incidence of grade 3 radiation pneumonitis and futility monitoring for efficacy. The results and conclusions of the study are not yet available.
