P2.05. Genomic Variants Associated with Response to Immune Checkpoint Inhibitors Amongst Asian Patients with Advanced NSCLC - PDF(Abstract)

Researchers at the National University Cancer Institute Singapore and Tan Tock Seng Hospital conducted a study to identify genomic variants associated with response to immune checkpoint inhibitors (ICIs) in Asian patients with advanced non-small cell lung cancer (NSCLC). The study enrolled 100 patients who were treated with ICIs between October 2013 and March 2022. The patients were genotyped and their response to ICIs was determined based on complete response, partial response, or sustained stable disease. The study found that 76% of patients were responders and 24% were non-responders. The median age of the patients was 64, and the majority were male, Chinese, and smokers. The study also found that patients who responded to ICIs had longer progression-free survival and overall survival compared to non-responders. <br /><br />The researchers performed a genome-wide association study (GWAS) and identified seven novel genetic biomarkers that were associated with ICI response in advanced NSCLC. These biomarkers included variants in the genes BMP6, IGFBP7-AS1, LINC02468, LINC02751, and LDLRAD4. The researchers believe that these gene signatures could be valuable tools in predicting ICI response in patients with advanced NSCLC. However, further validation in an independent patient cohort is needed. The study highlights the importance of genetic variation in determining drug response and suggests that genomic predictors could be used to personalize treatment decisions for NSCLC patients.
