P2.09. QT-interval Prolongation and Torsades de Pointes With EGFR TKIs in NSCLC: Targeted Literature Review - PDF(Abstract)

A targeted literature review was conducted to understand the risks of QT-interval prolongation (QTp) and torsades de pointes (TdP) associated with epidermal growth factor receptor tyrosine kinase inhibitors (EGFR TKIs) in patients with non-small cell lung cancer (NSCLC). The review identified 82 publications that met eligibility criteria and included a total of 17,401 patients with NSCLC. The incidence of QTp ranged from 0% to 27.8% in observational studies and from 0% to 11% in clinical trials. No deaths due to QTp or TdP were reported in the observational studies or clinical trials. However, five case reports described QTp events in patients receiving EGFR TKIs, and three case reports described TdP events, one of which resulted in death. <br /><br />The review suggests that approximately one third of the studies evaluating EGFR TKIs in NSCLC patients reported QTp events, but there were no deaths related to cardiotoxicity. Health care providers are advised to monitor patients for signs of cardiotoxicity and changes in heart rhythm. Future directions could include developing standardized definitions of QTp and improving monitoring methods for QTp and TdP in cancer patients. <br /><br />Overall, the targeted literature review highlights the importance of recognizing and treating QTp and TdP, serious conditions that can occur with some cancer treatments and may lead to sudden death if not addressed.
