P2.17. Characterizing Mechanisms of Treatment Resistance in SCLC - PDF(Abstract)

This abstract presents a study conducted to understand the mechanisms of treatment resistance in small cell lung cancer (SCLC) at a cellular level. The researchers performed single cell RNA sequencing (scRNA-seq) on tumor biopsies from five patients with SCLC, including one treatment-naive patient and four patients at relapse. The scRNA-seq data was analyzed to identify transcriptional differences between sensitive (S) and resistant (R) SCLC cells.<br /><br />The results showed significant transcriptional differences between S and R cells, particularly in pathways such as WNT and those governing non-neuroendocrine differentiation. R cells also had a higher degree of cell differentiation compared to treatment-naive and S cells. This increase in cell differentiation may represent a common underlying theme for some of the mechanisms of treatment resistance in SCLC.<br /><br />The researchers further validated their findings using scRNA-seq data from a different dataset consisting of 30,039 SCLC cells. The results from this dataset also showed a higher degree of cell differentiation in R cells compared to S cells.<br /><br />Overall, this study suggests that the mechanisms of treatment resistance in SCLC are heterogeneous, and an increase in cell differentiation may play a role in these mechanisms. Further studies are needed to better understand this phenomenon and validate these observations.<br /><br />The findings of this study provide valuable insights into the understanding of treatment resistance in SCLC and can potentially contribute to the development of novel therapies to improve patient outcomes.
