P2.20. A Precision Medicine Approach by Performing Comprehensive Cancer Genome Profiling of Thymomas and Recurrent Thymomas - PDF(Abstract)

This study aims to evaluate the molecular profiles of thymomas and recurrent thymomas, which are currently not well defined. The researchers conducted a pilot study on a selected group of patients with recurrent thymomas, comparing the biomolecular results obtained from both the primary and relapse samples. A total of 23 patients with recurrent thymomas were included in the study, with 40 surgical specimens available for analysis. The TruSight_Oncology (TSO500HT) kit was used to evaluate Tumour-Mutational-Burden (TMB), Microsatellite-Instability (MSI), and genetic variations in 523 genes. Next Generation Sequencing was performed on the specimens.<br /><br />The results showed that relevant molecular findings were observed in 83% of cases, mainly involving cell cycle control (74%) and DNA repair pathways (26%). No significant differences in the molecular profiles were found between primary and recurrent thymomas. Additionally, no significant associations were found between clinical variables and gene mutations. Based on the Next Generation Sequencing results, potential off-label treatments or participation in clinical trials could be suggested for over 50% of the cases.<br /><br />In conclusion, the study identified relevant molecular findings in both thymomas and recurrent thymomas, with no change in gene profile when the tumor relapses. By performing comprehensive cancer genome profiling, the researchers found multiple potential target therapies and active clinical trials.
