P2.21. A Novel Proteomic Technique Uncovers New Diagnostic Biomarkers for Malignant Pleural Mesothelioma - PDF(Abstract)

A study presented at the WCLC 2023 conference revealed a novel proteomic technique for identifying diagnostic biomarkers for malignant pleural mesothelioma (MPM). MPM is a highly aggressive cancer with a late diagnosis and poor prognosis. The study aimed to identify new biomarkers for MPM that could differentiate patients from control groups. <br /><br />A total of 177 plasma samples were collected from MPM patients, lung cancer patients, and asbestos-exposed individuals. The samples were processed using a unique method to enrich for tissue leakage proteins, and then analyzed by liquid chromatography-tandem mass spectrometry. Based on the analysis, 1006 unique proteins were identified, 539 of which were intracellular proteins. <br /><br />Using a random forest classifier and logistic regression model, the researchers identified four proteins that could differentiate MPM patients from asbestos-exposed controls with an accuracy of 0.887 and a negative predictive value of 0.978. Additionally, three proteins were identified that could differentiate lung cancer patients from asbestos-exposed controls with an accuracy of 0.873 and a negative predictive value of 0.899. <br /><br />Furthermore, the study found that the majority of intracellular proteins were upregulated in the MPM group compared to the control groups, suggesting their potential as biomarkers. Using the number of identified intracellular proteins alone, both the MPM group and the lung cancer group could be distinguished from the control group. <br /><br />Overall, this study identified new candidate diagnostic biomarkers for MPM, providing a less invasive alternative for diagnosis. However, further testing and validation is needed to confirm these results.
