Pdf Summary
The study conducted by researchers from the Department of Thoracic Surgery at Tangdu Hospital in Xi’an, China, focuses on gene therapy targeting the immune checkpoint CD155 in lung adenocarcinoma (LUAD), the most common subtype of lung cancer. Lung cancer remains a significant global health issue, contributing to a substantial percentage of cancer morbidity and mortality. CD155 is identified as a protein highly expressed in various tumors, playing roles in tumor progression and immune modulation within the tumor microenvironment.

The research involved evaluating CD155 expression in cancerous and adjacent tissues of 160 LUAD patients using immunohistochemical staining. The correlation of CD155 levels with patient survival was analyzed. Additionally, RNA sequencing and various cellular and molecular experiments were conducted to understand the functions and mechanisms linked to CD155.

Key findings suggest that CD155 promotes the proliferation and migration of LUAD cells. Higher CD155 expression is associated with poor prognosis for LUAD patients. Moreover, targeting CD155 through type 9 AAV (AAV9) to deliver the shCD155 sequence was found to be effective. The study demonstrated that AAV9-shCD155 could inhibit tumor growth and enhance the effectiveness of anti-PD-1 antibody therapy in humanized mice models.

The results indicate that downregulating CD155 expression in LUAD cells not only curtails their growth and motility but also potentially improves immune system infiltration and function, providing a promising avenue for improving LUAD treatment outcomes. This research was funded by the Shaanxi Outstanding Youth Natural Science Foundation and the National Natural Science Foundation of China, among others.
