PP01.49: FTIR Profiling of Serum Samples and its Association With Global DNA Methylation Percentage in Lung Cancer Patients

This study explores the potential of Fourier Transform Infrared Spectroscopy (FTIR) as a tool to detect global DNA methylation in lung cancer patients, utilizing its ability to analyze metabolites and nucleic acid structures. Conventional methods for detecting DNA methylation are often limited to research settings due to their complexity; hence, identifying simpler and cost-effective alternatives like FTIR is valuable.<br /><br />The study involved collecting peripheral blood and serum samples from 20 control participants and 6 lung cancer patients, all of whom were diagnosed with stage IV adenocarcinoma. Global DNA methylation levels were assessed using a colorimetric ELISA kit and FTIR spectroscopic analysis, with data correlated through multiple linear regression.<br /><br />Key findings indicate that lung cancer patients exhibit significant global DNA hypomethylation compared to the control group. This hypomethylation is observed regardless of gender and age, suggesting it as a characteristic feature of advanced lung cancer stages. Despite age-related changes in DNA methylation patterns generally, age did not significantly correlate with methylation levels in this study's population. <br /><br />The research supports the hypothesis that lung cancer alters DNA methylation patterns, specifically showing a decrease in global methylation. This characteristic hypomethylation could potentially be detected through peripheral blood analysis, providing a non-invasive diagnostic marker of the disease.<br /><br />The study emphasizes the need for further exploration of FTIR techniques for broad applicability in clinical diagnostics, making it a promising candidate for rapid and accessible lung cancer screening.
