Pdf Summary
The study focuses on the development of a novel diagnostic tool for lung cancer detection using Fe3O4 nanoparticles coated with polyethyleneimine (PEI) conjugated with DNA aptamers. Lung cancer, known for its aggressive nature, poses a significant challenge in early detection, as most diagnoses occur at advanced stages. The researchers utilized DNA aptamers, selected through Systematic Evolution of Ligands by Exponential Enrichment (SELEX), for their ability to selectively bind to lung cancer cells. The aptamers were tested against various lung cancer cell lines, including A549, HCC827, and H1299, to evaluate their specificity and binding affinity through flow cytometry.

In the experiments, a specific aptamer, Apt 7, demonstrated strong binding affinity to lung cancer cells, particularly the A549 cell line, whereas the control aptamer Apt RC9 did not. This was confirmed through various tests, including gel electrophoresis and immunofluorescence assays, which confirmed Apt 7's selective binding to cancer cells in both cultured cell models and clinical sputum samples from lung cancer patients.

The study reported a significant statistical difference in binding rates, with the nanoparticle-aptamer conjugate showing a 46% higher specific binding rate to HCC827 cells compared to unbound aptamers. Additionally, immunofluorescence assays performed on sputum samples from lung cancer patients indicated that Apt 7 could specifically identify tumor cells, thus showing promise for early lung cancer diagnostics.

This research indicates that the combination of PEI-coated Fe3O4 nanoparticles and selected aptamers offers a sensitive and specific method for detecting lung cancer cells, suggesting potential applications in early cancer detection and monitoring.
