Pdf Summary
The INHERIT Study investigates hereditary risk factors in thoracic cancers, specifically focusing on individuals with a family history of lung cancer and genetic predispositions. Its goal is to better understand these risks to inform lung cancer screening protocols.

Key objectives include identifying individuals with increased susceptibility to lung cancer through germline and somatic genetic testing and profiling tumor samples from predisposed persons. The study aims to analyze genetic variants using whole-genome sequencing to determine potential modifiers of known oncogenic mutations.

Participants are divided into three cohorts based on their genetic profile and family history:

- Cohort 1: Individuals with germline EGFR mutations known to be associated with lung cancer risk.

- Cohort 2: Individuals with non-EGFR germline mutations that may suggest inherited lung cancer risk, such as HER2, YAP1, and TP53 among others.

- Cohort 3: Individuals with a family history of lung cancer but without known pathogenic variants.

Eligibility for the study includes having a personal or family history of lung cancer and testing positive for or having a high risk of carrying specific genetic variants. Participants undergo genetic counseling, comprehensive family history documentation, and consent to provide research blood specimens.

The study offers various clinical follow-ups, including optional CT scans if clinically indicated. Results are communicated cautiously, especially if actionable, ensuring patients and their families understand their genetic predispositions. While only a portion of results is returned to participants, the study compiles a comprehensive repository of clinicopathological data to facilitate future research.

The trial is conducted across various states, with 179 participants currently enrolled, highlighting its broad reach and importance in genetic cancer research.
