This study investigates biomarkers associated with lung adenocarcinoma progression using proteomics and non-targeted metabolomics to address the challenges of early diagnosis and treatment of cancer metastasis, which often results in poor prognosis. The research highlights alterations in GOLT1B and PTGES3 genes and 27 related metabolites as significant markers linked to the metastasis of lung adenocarcinoma.<br /><br />Data were collected from 40 patients, categorized into groups based on metastasis status, using long-term follow-up data. Proteomic and metabolomic analyses revealed 99 differentially expressed proteins and 58 metabolites related to lung adenocarcinoma metastasis. In particular, GOLT1B and PTGES3 were identified as central genes strongly associated with metastasis.<br /><br />The study employed Pearson correlation to assess the relationship between clinical outcomes and differentially expressed proteins (DEPs) and metabolites (DEMs). A network diagram of these DEPs/DEMs was constructed using Cytoscape. The inhibitory impact of overexpressing GOLT1B and PTGES3 on tumor progression was tested through in vitro and in vivo experiments, suggesting these genes promote the degradation of HIF1-α, thereby exerting anti-tumor effects.<br /><br />Baseline characteristics compared between groups indicated no significant differences in age, gender, cancer stage, or smoking index, although a notable difference was observed in the status and progression-free survival (DFS).<br /><br />The research implies that GOLT1B and PTGES3 might suppress lung adenocarcinoma progression by influencing hypoxia-related pathways, offering potential new targets for therapeutic strategies. These findings could pave the way for earlier and more accurate diagnosis and improved treatment options, addressing current clinical challenges in managing lung adenocarcinoma metastasis.
