Pdf Summary
This study investigates the potential causal relationship between serum iron levels and lung cancer risk, highlighting the role of the HFE gene. It uses Mendelian Randomization (MR) and bioinformatics approaches to explore this association. Notably, the study identifies a significant single nucleotide polymorphism (SNP), rs1800562, related to the HFE gene, which is elevated in lung adenocarcinoma and squamous cell carcinoma. High expression of HFE correlates with poorer disease-specific survival in lung cancer patients and demonstrates various interactions with immune cell types and tumor-infiltrating lymphocytes (TILs) abundance in squamous cell lung cancer.

Elevated serum iron, ferritin, and transferrin saturation levels are linked to a reduced risk of squamous cell lung cancer, suggesting protective associations. The study utilizes data from the UK Biobank, applying statistical methods like fixed-effects inverse-variance weighted (IVW) model, MR-Egger, and weighted median methods for robust analysis. Furthermore, gene set enrichment and protein-protein interaction network analyses reveal multiple biological functions of HFE in lung cancer.

The research underscores HFE's potential influence on lung cancer progression through immune cell regulation and iron metabolism. It identifies two potential drugs that interact with HFE, suggesting therapeutic avenues. Overall, the findings propose that HFE plays a significant role in lung cancer development, emphasizing its involvement in immune regulation and iron handling in non-small cell lung cancer (NSCLC). This comprehensive study not only enhances understanding of lung cancer pathogenesis but also opens up new directions for targeted treatments.
