EP.02.11 Clinical and Biological Significance of Cystathionine Gamma-Lyase in Pulmonary Adenocarcinoma

This study investigates the clinical and biological significance of Cystathionine gamma-lyase (CTH) in pulmonary adenocarcinoma, a subtype of non-small cell lung cancer (NSCLC). CTH is an enzyme involved in producing endogenous hydrogen sulfide, influencing physiological processes like cell proliferation and migration. However, its role within the hypoxic tumor microenvironment of NSCLC is not well understood.<br /><br />Researchers analyzed tumor samples from 103 patients who underwent radical surgery for invasive pulmonary adenocarcinoma between 2017 and 2020. Using immunohistochemistry, they categorized CTH expression as high when over 10% of tumor cells stained positive. Over half (51.5%) of the tumors exhibited high CTH expression, which correlated significantly with microvascular invasion and tumor recurrence.<br /><br />Survival analysis showed that patients with high CTH expression had worse relapse-free and overall survival rates compared to those with lower expression levels, suggesting CTH as a potential prognostic marker. In vitro, knockdown of CTH in human pulmonary adenocarcinoma cell lines (ABC1 and VMRC-LCD) did not affect cell proliferation but significantly reduced tumor cell migration and invasion. Further investigation revealed that decreased CTH expression led to reduced levels of platelet-derived growth factor-AA (PDGF-AA), a growth factor associated with tumor cell motility.<br /><br />The study concludes that CTH enhances tumor cell motility through upregulating PDGF-AA, contributing to metastasis and recurrence in invasive pulmonary adenocarcinoma. Therefore, measuring CTH expression could serve as a predictive biomarker for tumor recurrence in patients with stage IA-IIIA disease, offering potential implications for prognosis and therapeutic strategies targeting tumor invasion and metastasis.
