EP.06.14 Decreased Expression of DAAM2 Is Correlated With Poor Prognosis and Tumor Immune Microenvironment in Lung Adenocarcinoma

This study investigated the role of Disheveled-associated activators of morphogenesis 2 (DAAM2) in lung adenocarcinoma (LUAD). Although DAAM2 has been implicated in tumorigenesis across various cancers, its function in LUAD remained unclear. Researchers analyzed DAAM2 expression using data from The Cancer Genome Atlas and TIMER2.0 databases, validated findings in a tissue microarray, and assessed the correlation with patient prognosis, clinicopathological features, and tumor immune microenvironment.<br /><br />The results revealed that DAAM2 expression is significantly downregulated in LUAD tissues, correlating with poorer overall survival, disease-specific survival, progression-free interval, and cumulative survival in patients. Functional enrichment suggested that DAAM2 may influence LUAD progression via pathways involved in the cell cycle, P53, Wnt, and TGF-β signaling. In vitro experiments demonstrated that overexpression of DAAM2 suppresses LUAD cell proliferation, migration, and invasion, indicating a tumor-suppressive role.<br /><br />Furthermore, DAAM2 expression levels were significantly associated with the infiltration of immune cells and markers within the tumor microenvironment, suggesting its involvement in modulating anti-tumor immunity. These findings propose that decreased DAAM2 expression contributes to LUAD progression by promoting tumor growth and altering the immune microenvironment.<br /><br />Overall, the study concludes that DAAM2 acts as a tumor suppressor in LUAD and holds potential as a prognostic biomarker and therapeutic target.
