EP.06.25 Analysis of LAT1 Protein Expression and Clinical Characteristics of Patients With Primary Lung Cancer

This study investigated the expression of L-type amino acid transporter 1 (LAT1), encoded by SLC7A5, in lung cancer and its association with prognosis and histological features. LAT1 facilitates the cellular uptake of large neutral amino acids and is known to be dysregulated in various cancers.<br /><br />The study analyzed 127 lung cancer patients treated at Kanazawa University Hospital between 2001 and 2024, using immunohistochemistry to evaluate LAT1 and other markers. LAT1 positivity rates were 58% in small cell carcinoma (SCLC), 50% in adenocarcinoma, and 94% in squamous cell carcinoma. Survival analysis showed significantly shorter disease-free survival (32.2 vs. 75.6 months, p=0.014, HR=2.087) and overall survival (p=0.024, HR=2.463) in LAT1-positive patients compared to LAT1-negative ones, indicating LAT1 expression as a negative prognostic factor.<br /><br />There was no significant correlation between LAT1 expression and molecular subtypes of SCLC. The study also examined LAT1 expression heterogeneity between primary and metastatic sites in 27 autopsy cases and found moderate correlation with an 84.2% concordance rate, suggesting primary tumor LAT1 status may reflect overall tumor burden.<br /><br />The findings align with previous reports in lung adenocarcinoma and biliary tract cancers, where LAT1 overexpression associates with poor outcomes. The authors propose that LAT1 contributes to cancer growth by promoting amino acid uptake. Given the limited heterogeneity across metastases, LAT1 is a promising prognostic biomarker and potential therapeutic target. Future development of LAT1-targeted therapies, including radioisotopes, is warranted to improve lung cancer management.
