EP.06.30 Single-Cell RNA Sequencing Reveals a Subset of FSIP1 Cancer Cells and Verified Its Value of Prognosis in Lung Adenocarcinoma

This study investigates the role of Fibrous Sheath-Interacting Protein 1 (FSIP1) in lung adenocarcinoma (LUAD), the most common subtype of non-small cell lung cancer (NSCLC). FSIP1, originally known as a component of sperm flagellum structure, has been implicated in promoting tumor growth, invasion, and drug resistance in various cancers, but its function and prognostic value in LUAD were previously unclear.<br /><br />Using single-cell RNA sequencing data from early-stage resectable LUAD samples, researchers identified a distinct cancer cell subset with high FSIP1 expression. Analysis of 180 LUAD tissue samples showed that elevated FSIP1 protein levels are significantly associated with poorer patient survival, confirmed via Kaplan-Meier analysis and Cox regression models. A prognostic nomogram incorporating FSIP1 expression effectively predicted patient outcomes, supported by Receiver Operating Characteristic (ROC) curves.<br /><br />Functional assays using FSIP1-silenced LUAD cell lines (A549 and H1975) demonstrated markedly reduced migration ability, as evidenced by wound-healing and Transwell migration assays. Western blot analyses revealed that FSIP1 knockdown suppresses the PI3K/AKT signaling pathway, a critical pathway involved in cancer progression.<br /><br />In summary, the study identifies FSIP1 as a novel prognostic biomarker for LUAD patients, where high expression correlates with worse survival. Furthermore, FSIP1 promotes LUAD cell migration and activates oncogenic PI3K/AKT signaling, suggesting it as a potential therapeutic target. These findings were supported by data from cell-based experiments and patient tissue analyses. The research was funded by health and science foundations in Hangzhou and Zhejiang, China, with no conflicts of interest declared.<br /><br />Presented at the IASLC World Conference on Lung Cancer 2025, this work highlights FSIP1’s significance in LUAD prognosis and its molecular role in tumor cell behavior.
