This study investigated the role of Disheveled-associated activators of morphogenesis 2 (DAAM2) in lung adenocarcinoma (LUAD), a common form of lung cancer. Using data from The Cancer Genome Atlas and TIMER2.0 databases, alongside tissue microarray validation, the researchers assessed DAAM2 expression in LUAD samples. They found that DAAM2 expression was significantly downregulated in LUAD tissues compared to normal tissues. Lower DAAM2 levels were associated with poorer overall survival, disease-specific survival, progression-free interval, and cumulative survival, indicating a correlation with unfavorable clinical outcomes.<br /><br />Functional enrichment analyses suggested that DAAM2 influences LUAD progression through key biological pathways, including the cell cycle, P53, Wnt, and TGF-β signaling pathways. Experimentally, overexpression of DAAM2 in LUAD cell lines inhibited cell proliferation, migration, and invasion, as confirmed by Cell Counting Kit-8 assay, cell scratch assay, and Transwell invasion assay. <br /><br />Moreover, the study revealed a significant association between DAAM2 expression and the tumor immune microenvironment, with variations in immune cell infiltration and immune markers correlating with DAAM2 levels. These findings suggest that DAAM2 not only acts as a tumor suppressor in LUAD but also affects immune dynamics within the tumor milieu.<br /><br />Overall, the results indicate that DAAM2 downregulation promotes LUAD progression, while its increased expression can inhibit tumor growth and spread. Therefore, DAAM2 holds potential as a prognostic biomarker and therapeutic target for LUAD, potentially guiding future treatment strategies aimed at improving patient outcomes.
